摘要
目的研究含α-β2-β1融合基因的重组菌株BL21(DE3)(pXETAB2B1)的免疫原性。方法用重组菌株BL21(DE3)(pXETAB2B1)制备包涵体和全细胞培养物两种抗原,采用氢氧化铝胶体作为免疫佐剂,免疫ICR小白鼠,用C型产气荚膜梭菌外毒素粗提物进行攻毒,观察小鼠的保护情况。结果包涵体加氢氧化铝凝胶制备的抗原保护效果对于1LD100和2LD100的攻毒均达到100%;全细胞培养物加氢氧化铝凝胶制备的抗原保护效果对于1LD100和2LD100的攻毒达到50%和20%。结论已获得具有一定免疫原性重组菌株,可作为预防C型产气荚膜梭菌所致疾病的候选菌株。
Objective To explore the immunogenicity of alpha toxin、beta 2 toxin and beta1 toxin fusion gene in the recombinant strain BL21(DE3)(pXETAB2BZ)of Clostridium perfringens type C.Methods Inclusion body and the strain culture of recombinant strain BL21(DE3)(pXETAB2BZ)were made and the aluminum hydroxide hydrate and Freund's adjuvant were added as adjuvant respectively.ICR mouse was immunized with different antigens and challenged with exotoxin of Clostridium perfringens type C.Immunogenicity of the recombinant strain were observed in mice.Results Immunization with inclusion body containing aluminum hydroxide gel could protect ICR mouse from the challenge of 1LD100 and 2LD100.The protective rate of the strain culture containing aluminum hydroxide gel was 70% and 20% from the challenge of 1LD100 and 2LD100 respectively;The inclusion body and the strain culture containing Freund's adjuvant was 50% and 10% from the challenge of 2LD100 of Clostridium perfingens C type toxin.Conclusion The recombinant strain BL21(DE3)(pXETAB2BZ)had immunogenicity and could be used as candidate strain of vaccination.
