摘要
目的建立小花棘豆中主体毒性物质野决明碱含量的HPLC测定方法。方法采用高效液相色谱法,色谱柱:ODS-C18(250mm×4.0mm,5μm);柱温为30℃.流动相A:甲醛,流速为0.4mL·min^-1;流动相B:0.5%柠檬酸钠+0.5%磷酸二氢钠,流速为0.6mL·min^-1;检测波长为254mm.结果野决明碱含量与对应的色谱峰面积在0.05~0.04mL·min^-1范围内具有良好的线性关系,r=0.9961;平均回收率为101.91%,精密度为RSD=0.42%(n=5).结论本方法可有效地用于测定小花棘豆中野决明碱的含量,是一种准确、灵敏、可靠的分析方法。
Objective To establish a method for the determination of the primary poisonous substance - thermopsine in oxytropis glabra DC by HPLC. Methods Analyses were carried out on ODS -C18 (250 mm ×4.0 mm, 5 μm) column. Mobile phase A: methanol, the flow rate 0.4 mL - rain-1 ; mobile phase B: 0.5% sodium citrate + 0.5% sodium dihydrogen phosphate, the flow rate 6 mL · min^-1 The column temperature was 30 ℃. The UV detection wavelength was set at 254 nm. Results The standard curve was linear in the range of 0. 005 - 0.04 mg · mL^-1, the corresponding correlation coefficient r = 0. 9961 ; the averaged recoveries was 101.91% and the precision RSD was 0.42% (n = 5). Conclusions The method is accurate, sensitive, and reliable. And it could be availably applied to determine thermopsine content in oxytropis glabra DC.
