摘要
目的探讨聚乙二醇-牛血红蛋白(polyethylene glycol-bovine hemoglobin,PEG-bHb)作为携氧剂加入St.Thomas心脏停搏液中对离体心脏的保护效果。方法将32只成年雄性SD大鼠按所使用的不同的心脏停搏液随机分为4组:St.Thomas液组(A组)、1∶2PEG-bHb组(B组)、1∶4PEG-bHb组(C组)和1∶8PEG-bHb组(D组),建立Langendorff离体心脏灌注模型,平衡20min后灌注冷心脏停搏液,30℃保存60min后恢复灌注,观察各组冠脉流出液的心肌肌钙蛋白I(cTnI)含量和心肌三磷酸腺苷(ATP)含量,并做病理检查。结果恢复灌注后,B组、C组、D组的cTnI均低于A组(F=52.955,P<0.05),ATP含量均高于A组(F=68.757,P<0.05),心肌超微结构得到较好的保护。B组、C组心肌含水量低于A组(F=3.048,P<0.05)。结论与St.Thomas液相比,以PEG-bHb制成的含血心脏停搏液在离体鼠心缺血期间能够提供更好的心肌保护效果。
Objective To investigate the effect of polyethylene glycol-bovine hemoglobin (PEG-bHb), which was used as an oxygen carrier in eardioplegie solution, on the protection of isolated rat hearts. Methods The hearts of 32 male SD rats were harvested and transferred to Langendorff circuit. They were divided into 4 groups according to eardioeplegia.. St. Thomas group (group A), 1 :2 PEG-bHb group (group B), 1 : 4 PEG-bHb group (group C) and 1 : 8 PEG-bHb group (group D). After 20rain balance period, hearts were perfused with cold (4C) cardioplegic solutions, and preserved at 30'C for 60min, then reperfused. Levels of cardiac troponin I (eTn l) and adenosine triphosphate (ATP) contant in coronary effuent were detected, and ultrastruetures of myoeardium were observed. Results After reperfusion, eTn I contant of group A were higher (F=52. 955, P〈0.05) and ATP eontant were lower (F=68. 757, P〈0.05) than those in group B, group C and group D. Myocardial water eontant were lower in group B and group C(F=3. 048, P〈0. 05). Conclusion PEG-bHb in eardioplegie solutions can provide better myocardial protection during isehemia.
