甲钴胺对SD大鼠坐骨神经损害后背根神经元的保护作用 被引量:8

摘要 目的:观察甲钴胺对坐骨神经损害后背根神经元病理形态改变的影响。方法:将30只SD大鼠分为正常对照组、模型组和甲钴胺组,每组各10只。以大鼠坐骨神经建立周围神经损害动物模型,以背根神经节病理切片(光镜、电镜)中的神经元变化情况为观察指标,考察甲钴胺对坐骨神经损害后神经元的影响。结果:4周后模型组神经元坏死率为(60.28±3.56)%,而甲钴胺组为(11.66±1.52)%,甲钴胺组明显低于模型组(P<0.01)。模型组4周时可见胞质、胞核大量空泡变性,内质网水肿明显,染色质聚集成块,分布不均,而甲钴胺组电镜观察胞膜、核膜比较完整,细胞器分布均匀,内质网无明显肿胀,总体观与正常对照组相似。结论:甲钴胺对坐骨神经损害后神经元有确切的保护作用。 Objective: To observe the protective effect of methycobal on the neuron of damaged sciatic nerve in rats. Methods: Thirty SD rats were divided into normal control group, model group, and methycobal group with 10 rats in each group. Sciatic nerve lesion model was established in rats, and the morphological changes of neuron ceils of DRG treated with methycobal were observed by light and electron microscopy. Results: The rate of neuron necrosis in model group was(60.28± 3.56)% after 4 weeks, and it was ( 11.66±1.52)% in methycobal group. The rate of neuron necrosis in model group was significandy lower than that in model group(P 〈 0.01 ). There were mass of vacuolar degeneration in cypoplasm and nucleus, significant ER edema, and aggretated caryotin in model group at the forth week. In methycobal group, cell membrane and caryotheca integrated relatively, cell organs distributed without ER edema, and the overall view was similar with the normal control group under the electron microscopy. Conclusion: Methycobal shows protective effect on neuron damaged sciatic nerve.
