摘要
目的探讨神经保护肽慢病毒(rLent/NT4-NAP)对老年性痴呆(AD)大鼠海马神经元的保护作用。方法选择无菌级雄性Wister大鼠40只,随机分为对照组,假手术组,AD组,rLent/NT4-NAP组,每组10只。建立β-淀粉样蛋白(Aβ)大鼠海马注射的实验性AD大鼠动物模型[2],利用水迷宫试验检测大鼠记忆能力,组织切片HE染色观察大鼠海马神经元变化,免疫组织化学染色观察caspase-3的表达。结果与AD组相比,rLent/NT4-NAP组水迷宫试验逃避潜伏期明显缩短,错误次数显著降低(P<0.01);HE染色可见rLent/NT4-NAP组大鼠海马损伤神经元数量较AD组少;免疫组化:rLent/NT4-NAP组大鼠海马区caspase-3阳性细胞数较AD组明显减少,但较对照组caspase-3阳性细胞数增加。结论NAP对老年性痴呆大鼠海马神经元具有保护作用。
Objective To investigate the neuron protective effect of Lentivirus neuroprotective peptid to Alzheimer's disease mice in the hippocampus.Methods male Wister mice were randomly assigned into sham operation grop with 10 mice,control group with 10 mice,AD grop with 10 mice,rLent/NT4-NAP grop with 10 mice. Beta-amyloid protein injection into mice hippoe21mpus were applied to make experimental mice model of AD. The ability of learning and memory was tested by the water maze, the neuron changes of hippocampus were objected by HE drum of tissue section,caspase-3 expressings were objected by immunohistochemistry. Results the escape eclipse period of rLent/NT4-NAP grop is less than that ofAD group(P 〈 0.01). Damage neuron of rLent/NT4-NAP grop are less than that of AD group. Conclusion treatment of NAP can protect neuron of hippocampus in the Alzheimer's disease mice.
