细胞粘附肽(RGDS)抑制人胃癌细胞BGC823生长及诱导其凋亡

摘要 目的研究细胞粘附肽(RGDS)对人胃癌细胞系BGC823(Human stomach cancer BGC823)生长增殖的影响,进而探讨RGDS诱导BGC823细胞凋亡的作用。方法用不同浓度的RGDS处理BGC823细胞48h后,MTT法检测RGDS对BGC823细胞生长增殖的抑制作用,通过HE染色、透射电子显微镜、免疫组织化学、流式细胞术和凋亡细胞的DNA片断等方法观察凋亡细胞的形态结构变化以及定性、定量检测细胞凋亡。结果RGDS能抑制BGC823细胞的生长增殖,并呈浓度依赖性;RGDS诱导BGC823细胞凋亡的作用机制可能通过抑制凋亡抑制蛋白Survivin的表达引发这一途径。结论RGDS能够通过诱导BGC823细胞凋亡,从而抑制肿瘤细胞的增殖。 Objective To investigate the effect of cellular adhesion motif RGDS on the growth and proliferation of human stomach cancer cells BGC823, and to study the function of cellular adhesion motif RGDS in inducing BGC823 apoptosis. Methods BGC823 cells were treated with cellular adhesion motif RGDS at various concentrations. MTT assay was used to examine the growth and proliferation of BGC823 cells after treament with cellular adhesion motif RGDS for 48 hours. HE staining and electromicroscope were used to observe the morphology of apoptotic cells, and Survivin, flow cytornetry were also used to analyze the BGC823 apoptosis. Results cellular adhesion motif RGDS significantly inhibits the growth and proliferation of BGC823 ceils in dose-dependent manner. And cellular adhesion motif RGDS also induce the apoptosis of BGC823 cells. Inducing apoptosis was caused by inhibiting the expression of Survivin. Conclusion cellular adhesion motif RGDS may inhibit turnout cell growth and proliferation through inducing apoptosis.
