摘要
目的:观察原代培养的小鼠乳鼠心肌细胞的形态特征。方法:Ⅱ型胶原酶、胰蛋白酶消化心脏组织,差速贴壁分离法。结果:12h心肌细胞基本贴壁,第2天可长成单层,第3~4天细胞伸出伪足相互接触交织成网,逐渐形成细胞簇或细胞单层,搏动呈同步性,收缩明显有力,搏动频率为30~120次/min。结论:用Ⅱ型胶原酶、胰蛋白酶分离培养小鼠乳鼠心肌细胞可获得形态、活力良好的心肌细胞。
Objective: To Observe the morphological characteristics of the cultured cardiomyocytes. Methods: The hearts were digested by collagenase type Ⅱ , trypsin and D Nase Ⅰ. The cell suspension was preplated to reduce the fibroblast content. Results: The cells spread over the first day and developed their characteristic morphology by day 3 or 4. They were arranged in loose and interlacing networks of single cells. The cells beat spontaneously at 30-110 per minute. Conclusion: It can obtain cardiomyocytes with well morphology and spontaneous beating with collagenase type Ⅱ , trypsinandDNase Ⅰ.
