MMP-9、VEGF和PTEN在乳腺癌中的表达及其临床意义 被引量:7

摘要 目的:研究基质金属蛋白酶-9(MMP-9)、血管内皮生长因子(VEGF)和第10号染色体同源丢失性磷酸酶-张力蛋白基因(PTEN)在乳腺癌中的表达,探讨其在乳腺癌发生、发展过程中的临床意义。方法:运用免疫组织化学Envision法检测56例乳腺癌和22例乳腺良性病变组织中MMP-9、VEGF和PTEN的表达情况,并分析其与临床病理参数的关系。结果:(1)MMP-9和VEGF在乳腺癌中阳性表达率分别为69.64%(39/56)、78.57%(44/56),在乳腺良性病变组织中分别为45.45%(10/22)、36.36%(8/22),差异有统计学意义(P<0.05);PTEN在乳腺癌中表达率为46.43%(26/56),在乳腺良性病变组织中为86.36%(19/22),差异有统计学意义(P<0.01)。(2)MMP-9和VEGF的表达均与组织学分级和腋窝淋巴结转移呈正相关(P<0.05)。PTEN蛋白的表达与腋窝淋巴结转移呈负相关(P<0.05),而与组织学分级无关(P>0.05)。(3)MMP-9和VEGF在乳腺癌中的表达呈正相关(P<0.05)。结论:MMP-9和VEGF参与乳腺癌的发生、发展过程,可作为评估乳腺癌浸润和转移的重要生物学指标,PTEN蛋白表达异常,可能与乳腺癌的发生、发展和预后有关。 Objective: To study expression of matrix metalloproteinase-9 (MMP-9), vascular endothelial growth factor(VEGF) and phosphatase and tensin homolog deleted from chromosome 10(PTEN) in breast cancer and evaluate its significance to clinicopathology of this tumor invasion and metastasis. Methods: The immunohistochemical staining method of Envision was used to evaluate the expression of MMP-9, VEGF and PTEN in 56 cases of breast cancer and 22 cases of benign breast diseases and its correlations to clinicopathologic features were analyzed. Results: (1) The positive incidence of MMP-9 and VEGF expression in 56 cases of breast cancer were 69. 64%(39/56)and 78. 57% (44/56) respectively,which was improved compared with the expression in benign breast diseases were 45.45%and 36.36% (P 〈0.05). The higher level expression of PTEN in 56 cases with the rate 46.43%(26/56) ,which was reduced compared with the expression in benign breast diseases was 86.36% (P 〈0.01). (2)The expression of MMP-9 and VEGF was positively correlated to axillary lymph node metastasis and pathological stage (P〈0.05). The expression of PTEN was negatively correlated to axillary lymph node metastasis (P〈0.05. On the contrary, there was no significant correlation between PTEN expression and pathological stage (P 〉0.05). (3) There was a positive correlation between the expression of MMP-9 and that of VEGF in breast cancer tissue (P〈0.05). Conclusions: MMP-9 was associated with VEGF in breast cancer, and could serve as an important index in the aspect of invasiveness and metastasis in breast cancer. It showed that tumor suppressor PTEN gene was abnormally, it may play roles in pathogenesis, development and prognosis in breast cancers.
