摘要
目的利用人前列腺癌裸鼠骨转移模型筛选高骨转移潜能的细胞亚株,并分析其细胞生物学特性。方法采用人前列腺癌细胞系PC-3左心室注射法制作裸鼠骨转移模型,反复取骨转移灶肿瘤细胞体外培养和造模,筛选高骨转移倾向的细胞亚株。结果筛选出具有高骨转移潜能的细胞亚株T3B,骨转移率为80%,显著高于母系PC-3骨转移率(P<0.05);其细胞生物学特性与母系PC-3比较,其体外黏附和侵袭能力极显著增高(P<0.01),而细胞增殖率、软琼脂克隆形成率、裸鼠皮下成瘤率及瘤体大小的差别无显著性意义(P>0.05)。结论运用裸鼠骨转移模型筛选,获得1株高骨转移潜能的人前列腺癌PC-3细胞系亚株。
Objective To select a human prostate carcinoma subpopulation with highly metastatic potential to bone in nude mice by animal models of bone metastases and study its biological characteristics. Methods Human prostate carcinoma cell line PC-3 cells were injected into the left cardiac ventricle of nude mice to generate bone metastases in vivo, then PC-3 cell subpopulations were screened out by serial recovery from bone metastasis and inoculation in vitro. Results A subpopulation of PC-3 was obtained by a second round of selection and denominated T3B. Compared to parental PC-3 (40%), T3B showed a higher metastatic potential to bone(80 %) in vivo (P〈0. 05), and presented a significant increase in the adhesion and invasion ability in vitro (P〈0. 01 ), but no significant difference in the soft-agar colony formation rate, tumorigenicity and tumor growth rate(P〉0. 05). Conclusion A human prostate carcinoma cell subpopulation with highly metastatic potential to bone is obtained in vivo.
