摘要
目的:探讨长托宁(Penehyclidinehydrochloride,PHCD)对感染性脑水肿保护作用。方法:雄性S-D大鼠105只,200~220g,随机分为5组(n=21只):对照组(C组)、水肿组(L组)、长托宁低、中、高剂量组(DL组0.05mg/kg、DM组0.15mg/kg和DH组0.45mg/kg)。向颈内动脉注射脂多糖(Lipopolysaccharide,LPS)150μg(0.15ml)建立大鼠感染性脑水肿模型。C组和L组腹腔注射生理盐水1ml,10min后向左颈内动脉注入无菌生理盐水0.15ml或LPS0.15ml;DL组、DM组DH组在注射LPS10min前分别腹腔给予0.05mg/kg、0.15mg/kg、0.45mg/kg长托宁。每组分别在手术后4h、6h和12h随机处死7只大鼠,每个时间点的5只用于测定脑组织含水量变化和光镜观察,其余2只电镜观察脑组织病理形态改变。结果:与C组相比,L组、DL组、DM组和DH组各时间点脑含水量增加(P<0.05);与L组比较,DL组脑含水量改变不明显(P>0.05),DM组和DH组脑含水量明显减少(P<0.05)。DM组和DH组病理学损伤较L组轻。结论:中高剂量长托宁预先给药可明显减轻脑水肿,改善血管内皮和神经元的损伤,对感染性脑水肿有治疗作用。
Objective:To investigate the effects of Penehyclidinehydrochloride(PH(:D) on endotoxin-induced cerebral edema. Methods : one handred and five S-D male rats ( 200 - 220g )were randomly divided into 5 groups (n=21 ) : groupC control ; groupL LPS;group DL,DM and DH received intraperitoneal PHCD 0.05,0.15 or 0.45mg/kg 10rain before lipopolysaccharide(LPS) administration. Cerebral edema was induced by internal carotid arterial LPS 150μg. Seven animals each group were decapitated at 4,5 and 12h after operation and their brains were immediately removed for determination of water content of brain and microscopic examination. Results:Administration of LPS caused severe brain damage and significantly increased water content of the brain.The LPS-induced changes were mitigated by pretreatment with different doses of PHCD in groupDM and DH,but there was no significant difference between the two groups. Conclusion:Pretreatment with PHCD can attenuate LPS-induced cerebral edema.
