摘要
目的建立胃癌的原位模型,研究葡萄球菌肠毒素B(SEB)对实验性原位接种胃癌的抑瘤作用。方法采用人胃癌细胞株SCG-7901,27只裸鼠和SEB。将SCG-7901接种于裸鼠皮下,形成实体瘤。取出后分割成小块,再接种在裸鼠胃壁浆膜下。2周后行SEB治疗。观察原位移植瘤在模型组和治疗组的生长及侵袭转移的病理变化。结果①胃浆膜下原位接种肿瘤在2周后可达0.8 cm3。②SEB可大面积杀伤肿瘤细胞,但同时对周边非癌组织亦有破坏,并可导致肿瘤细胞的脱落、种植。结论成功建立了胃癌原位模型,可为研究胃癌的侵袭转移提供与人胃癌相近的微环境。SEB具有很强的杀伤肿瘤细胞的作用,无淋巴道和血道的转移;但对周边非癌组织也有一定的破坏,并由于肿瘤细胞的坏死、脱落,可导致肿瘤细胞的种植。
Objective To investigate anti-tumor effect of SEB by orthotopic transplanted mouse model of gastric carcinoma. Methods Human gastric carcinoma cell strains SCG-7901 were inoculated subcutaneously into 27 nude mice. When solid tumor formed, the tumors were harvested and cut into pieces, then inoculated in subserosa of gastric wall of the mice. The mice were treated with SEB 2 weeks later. The pathological changes metabasis of orthotopic transplanted tumors in the mice were observed. Results (1)Two weeks after the gastric inoculation, orthotopic transplanted tumors were 0. 8 cm^3;(2)SEB could cause tumor cells killed greatly, although it could destroy the non-cancer tissue around carcinoma, and even lead tumor cells shedding and implantation. Conclusion The orthotopic transplanted mice model of gastric carcinoma is established successly. Inoculation of tumor tablet in subserosa of gastric wall can provide microenvironment, which is familiar with human gastric cancer, to investigate the invasion and metastasis of gastric cancer. SEB may cause gastric tumor killed. No invasion of lymphatic vessel and blood vessel can be observed,though SEB can devastate the non-cancer tissue around carcinoma, because of necrosis and amotio of tumor cell, and even lead tumorcell implantation.
