摘要
目的通过运用暴发性肝功能衰竭大鼠模型,从微生态学的角度探讨茵陈合剂对肝脏细胞再生的影响。方法将60只健康Wistar大鼠随机分为5组,设一组为健康对照组,其余用D-氨基半乳糖按2 g/kg浓度腹腔注射造模,分成不干预组、贝飞达组、纳米茵陈合剂组、贝飞达联合茵陈合剂治疗组,于14 d及28 d动态观察各组大鼠肠道菌群、血清谷丙转氨酶(ALT)、总胆红素(TBIL)的变化,血清肿瘤坏死因子(TNF-α)及肝细胞生长因子(HGF)的变化,门静脉血清内毒素(LPS)的变化及肝脏细胞的凋亡情况。结果D-氨基半乳糖(D-Gal)造模后,不干预组的各项指标同健康对照组比较有显著差异(P<0.01)。贝飞达组、纳米茵陈合剂组、贝飞达联合茵陈合剂治疗组14 d天及28 d两次检测各项指标结果,同不干预组间比较,有显著差异(P<0.01),各治疗组14 d及28 d两次检测各项指标结果组内进行比较,有显著差异(P<0.01),不干预组内进行比较P>0.05,无统计学意义。结论茵陈合剂通过促进双歧杆菌等益生菌的生长,抑制肝脏细菌易位,抑制G-杆菌生长,有效控制内毒素血症,切断FHF造成肝脏细胞损伤的关键环节,降低TNF-α分泌,促进HGF的表达,抑制肝细胞的凋亡,为肝细胞的再生提供了有利的条件。
Objective To study the effects of YINCHEN mixture on hepatic cells regeneration in fulminant hepatic failure rats from mieroeeologieal point of view. Methods Sixty healthy Wistar rats were randomly divided into 5 groups: normal control group, model group, BeiFeiDa group, YinChen mixture group, BeiFeiDa and YinChen mixture group. All rats except the normal control group were injected intra- peritoneally with D-gal 2 g/kg to induce fulminant hepatic failure. The changes of indexes such as intestinal mieroflora ; ALT,TIBI,TNF-α, HGF, LPS in the vena portalis,liver cells apoptosis were observed dynamically. Results There were significant differences in all the testing indexes between model group and normal control group( P 〈 0.01 ). And the significant differences of the testing indexes measured at various time points ( 14 days and 28 days after drugs were given) in rats were also observed when comparing BeiFeiDa group, YinChen mixture group, BeiFeiDa and YinChen mixture group with model group (P 〈 0.01 ). There were also statistically significant differences in each therapeutic group at various time points (P 〈 0.01 ). Conclusion By fostering bifid bacterium,YinChen mixture can depress the bacterial translocation in rats with fulminant hepatic failure and the growing of G-bacillus, control endoxemia, cut down the key factor of hepatic cell injury, degrade the secretion of TNF-α, enhance HGF expression, inhibit the apoptosis of liver cells and provide favorable condition for the regeneration of hepatic cells .
