摘要
目的探讨gefarnate滴眼液对碱烧伤兔结膜杯状细胞密度及粘蛋白分泌功能的影响。方法新西兰白兔20只(40只眼),制作碱烧伤干眼模型,随机分为对照组和实验组,分别滴生理盐水和gefarnate滴眼液,术后7、14、21、28d分别行结膜印迹细胞学检查;7d、28d各取结膜进行免疫组织化学检测。结果实验组杯状细胞密度在术后1个月内逐步上调,对照组未见明显的上调作用,两组差异有统计学意义(P<0.05);实验组MUC5AC表达率和积分在术后7d和28d均比对照组明显增高,差异有统计学意义(P<0.05)。结论Gefarnate滴眼液可明显促进杯状细胞增生和粘蛋白MUC5AC的分泌,暗示其在干眼治疗方面具有一定的应用前景。
Objective The research showed that abnormality of goblet cell and mucin in conjunctiva results in dry eyes.How to improve the goblet cell to secret mucin is very important for the management of dry eye.Present study was to discuss the effect of gefarnate drops on secretion of mucin and density of goblet cell in rabbit conjunctiva with alkali injury.Methods The dry eye model was established by putting the filter paper with 1.0 mol/L NaOH on the conjunctiva at 2 mm superior to cornea for 90 seconds in 40 eyes of 20 New Zealand rabbits and then divided into two groups randomly.Gefarnate drops was utilized topically 3 times per day for 28 days in other 20 model eyes as experimental group and normal saline solution was used at a same way in other 20 model eyes as control group.Impression cytology staining was performed in 7,14,21,28 days after operation respectively for the detect of goblet cell density.Conjunctival MUC5AC was determined by immunohistochemical assay in 7 and 28 days after modeling.Results No goblet cell was detected in burning conjunctiva in the 7th,14th and 21st day and a few goblet cells were found in the 28th day after modeling in the control group.However,the densities of goblet cells in experiment group was gradually ascended with time increase after operation in experimental group,showing a statistically significant difference in the 28th day between experimental group and control group(t=38.263,P〈0.01).No MUC5AC was expressed and was(3.70±2.26)% in the 28th day in the control group,and in the experimental group,the expression rate reached(5.15±2.45)% in the 7th day and(18.20±1.33)% in the 28th day,indicating a significant increase in experimental group.The score of MUC5AC was higher in 7 days,28 days after operation in experimental group than that of control group,having a statistically significant difference in comparison with control group(t=76.321,P〈0.01).Conclusion Gefarnate drops promote proliferation of goblet cell and secretion of mucin MUC5AC.These results suggest that gefarnate drops is available to the treatment of dry eye.
