芍芪多苷对胶原性关节炎大鼠单核巨噬细胞的影响 被引量:2

The effects of SQDG (芍芪多苷) on monocyte-macrophage from rats with collagen-induced arthritis

摘要 目的:研究芍芪多苷(SQDG)在炎症不同时期对胶原性关节炎(collagen-induced arthritis,CIA)大鼠单核巨噬细胞功能的影响。方法:建立鸡Ⅱ型胶原诱导的大鼠关节炎模型,Ficoll密度梯度离心法分离外周血单核细胞(peripheral blood mononuclear cell,PBMC);贴壁法分离PBMC中的单核巨噬细胞(monocyte/macrophage,Mo/Mφ),吞噬中性红方法检测Mo/Mφ的吞噬功能;小鼠淋巴细胞增殖法检测IL-1活性;MTT法检测T淋巴细胞增殖功能。结果:SQDG(21.25、42.5、85mg/kg)ig给药能增强正常大鼠Mo/Mφ的吞噬功能;抑制CIA大鼠过强的Mo/Mφ的吞噬功能,并使其分泌增高的IL-1水平降至正常范围内;体外试验SQDG(10、20、40、80、160mg/L)使正常大鼠的Mo/Mφ的吞噬功能增强,对IL-1的产生无影响;SQDG作用后的Mo/Mφ对T淋巴细胞增殖功能无明显影响。d7体外给予SQDG可使CIA大鼠增强的Mo/Mφ的吞噬功能降低;分泌升高的IL-1水平降低;和SQDG作用后的Mo/Mφ对T淋巴细胞增殖功能无明显影响;d15、d23体外给予SQDG可使CIA大鼠Mo/Mφ明显增强的吞噬功能降低;升高的IL-1水平降低;和SQDG作用后的Mo/Mφ使增强的T淋巴细胞增殖反应降低。结论:SQDG可能通过调节单核巨噬细胞的功能,调节CIA大鼠异常的免疫功能。 Aim :To study the effects of SQDG(芍芪多苷) on macrophage from rats with collagen-induced arthritis (CIA) . Methods: Chicken type 11 collagen was used to induce CIA in rats, applied Ficoll density gradient centrifugal method to separate peripheral blood mononuclear cell(PBMC) , adherence method to obtain monocyte/macrophage( Mo/Mφ), through the experiment of neutral red phagocytosis detection the phagocytosis function of PBMC ; IL-1 activity was measured by thymocyte proliferation assay ; T lymphocyte cells proliferation was determined by 3-(4,5-dimethylthiazal-2yl) 2,5- diphenyltetrazoli- umbromide(MTT) assay. Results: d14 ig SQDG (21.25,42.5,85mg/kg) could enhance phagocytosis function of Mo/Mφ of normal rats and suppress the augmented phagocytosis function of Mo/Mφ of CIA rats , cut down over-production of IL-1 to normal level. Administration SQDG (10,20,40,80,160mg/L) in vitro could enhance the phagocytosis function of Mo/Mφ of normal rats ,didnt affect the production of IL-1 ,the proliferation of T lymphocyte cells from peripheral blood co-cultured with the Mo/Mφ which had been co-cultured with SQDG didnt be affected in normal rats. D7 administration SQDG in vitro could cut down the phagocytosis function of Mo/Mφ and over-production of IL-1 of CIA rats ,however it had little influence to the proliferation of T lymphocyte cells cocultured with Mo/Mφ which had been co-cultured with SQDG; d15 ,d23 SQDG could cut down the phagocytosis function of Mo/Mφ and overproduction of IL-1 of CIA. Rats ; and could suppress the proliferation of T lymphocyte cells co-cultured with Mo/Mφ which had been co-cultured with SQDG. Conclusion: SQDG may has an improvement function on abnormal immunity through regulated the fuction of mononuclear macrophage cells .
