摘要
目的:探讨缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)和基质金属蛋白酶-7(matrix metal oproteinase-7,MMP-7)在直肠癌组织中的表达及相关性,并评价二者在直肠癌进展中的作用。方法:采用免疫组织化学SP法检测52例直肠癌组织中HIF-1α和MMP-7蛋白表达情况,分析二者的相关性及与临床病理特征的关系。结果:直肠癌组织中HIF-1α蛋白阳性表达率为67.3%,与对照组相比差异有统计学意义(P<0.01),且与淋巴结转移、Dukes分期情况相关(P<0.01)。MMP-7蛋白在直肠癌组织中阳性表达率为71.2%,显著高于对照组(P<0.01),且与淋巴结转移和Dukes分期情况也相关(P<0.05)。HIF-1α与MMP-7呈正相关(r=0.478,P<0.01)。结论:HIF-1α和MMP-7在直肠癌的进展中起重要作用,HIF-1α可能通过调节MMP-7的过度表达参与直肠癌的发生发展。
Objective: To study the expressions and the relation of Hypoxia inducible factor-1 α (HIF-1 α) and Matrix metal oproteinase-7 (MMP-7) in rectal cancer and evaluate their roles in rectal carcinogenesis and development. Methods: The expressions of HIF-1α and MMP-7 in 52 cases of rectal cancer was detected by immunohistochemical SP technique. Re. suits: The expression of HIF-1α in rectal carcinoma (67.3 %) was higher than that in control group (P〈0.01). The positive rate of HIF-1α was closely related with local lymph node metastasis and Dukes stage (P〈0. 01). The expression of MMP-7 in rectal carcinoma (71.2 %) was much higher than in the control group (P〈0. 01) and was closely related with local lymph mode metastasis and Dukes stage (P〈0.05). The expression of HIF-1 a was positively correlated with MMP-7 protein (r=0.478,P〈0.01). Conclusion HIF-1α and MMP-7 play important roles in the evolvement of rectal carcinoma, HIF-1α may participate the occurrence and development of rectal carcinoma by regulating the excess expression of MMP-7.
