FDP对微波辐射后大鼠海马COX表达的影响 被引量:3

Effects of fructose-l, 6-diphosphate on COX expression of mitochondrial respiratory chain in rats' hippocampus after microwave exposure

摘要 目的 探讨1,6-二磷酸果糖(FDP)对微波辐射后大鼠海马线粒体呼吸链相关基因细胞色素C氧化酶(cytochrome c oxidase,COX)表达的影响。方法 Wistar雄性大鼠随机分为假辐射组(5只),辐射组(10只)和辐射+药物组(10只)。采用30mW/cm^2微波辐射,辐射+药物组于辐射前3d腹腔注射350mg/kg FDP,连续给药3或10d,1次,d。于辐射后6h和7d活杀取海马,采用反转录-聚合酶链反应(RT-PCR)和图像分析检测大鼠海马组织COX亚基Ⅰ、Ⅱ和Ⅳ mRNA表达,通过Western blot和图像分析检测大鼠海马组织COXI蛋白表达变化。结果 辐射后6h和7d大鼠海马COX Ⅰ、Ⅱ和Ⅳ mRNA表达均较假辐射组降低,药物组大鼠海马COXⅠ、Ⅱ和 Ⅳ mRNA与相应辐射组相比均见表达增加,以给药后3d(辐射后6h)组增加最为显著(tcox Ⅰ=3.2205,tcox Ⅱ=3.5762,tcox Ⅳ=3.0408,P〈0.05)。辐射后6h和7d大鼠海马COX Ⅰ蛋白表达均较假辐射组减少,给药后3d(辐射后6h)与辐射后6h相比COXI蛋白表达升高(t=2.9614,P〈0.05),给药后10d(辐射后7d)与辐射后7d相比改变不显著。结论FDP对微波辐射后大鼠海马线粒体呼吸链相关基因COX表达降低有一定的促恢复作用。 Objective To investigate the effects of fructose-1,6-diphosphate (FDP) on the gene COX expression of mitochondrial respiratory chain in rats' hippocampus after microwave exposure. Methods 25 male rats were exposed to microwave that the average power density was 30 mW/cm^2. FDP was injected intraperitoneally once per day 3 d before exposure, lasting 3 ds or 10 ds. Rats were sacrificed at 6 h and 7 d after exposure. Reverse transcription-polymerase chain reaction (RT-PCR) and image analysis were used to study the expression of COX Ⅰ , Ⅱ and Ⅳ mRNA. The COX Ⅰ protein was detected by Western blot and image analysis. Results The expression level of COX Ⅰ , Ⅱ and Ⅳ mRNA were decreased at 6 h and 7 d after exposure while in administration group the expression level increased especially at 3 d after administration (tcox Ⅰ - 3.2205, tcox Ⅱ- 3.5762, tcox Ⅳ = 3.0408, P 〈 0.05). The COX Ⅰ. protein level decreased after exposure while it increased at 3 d after administration (t = 2.9614, P 〈 0.05), but there was no significant change at 10 d after administration. Conclusions FDP could recover the decrease of COX expression level of mitochondrial respiratory chain in rats' hippocampus after microwave exposure.
