CO_2气腹对人宫颈癌CasKi细胞p-ERK和p-Akt表达的影响 被引量:1

摘要 目的:探讨CO2气腹对人宫颈癌CasKi细胞信号转导通路的影响。方法:建立腹腔镜CO2气腹体外模型,以不同压力(12mmHg、13mmHg及14mmHg)的CO2气腹对人宫颈癌CasKi细胞处理2h,设未处理组为对照组。采用四唑盐比色试验(MTT)、免疫细胞化学法、Western blotting检测CasKi细胞体外生长情况、相关信号转导通路核心蛋白p-ERK和p-Akt的表达水平。结果:CO2处理组中CasKi细胞增殖加快,相关信号转导通路核心蛋白p-ERK和p-Akt的表达显著增加(P<0.01)。各个CO2处理组间,CasKi细胞增殖及p-ERK与p-Akt的表达无显著差异(P>0.05)。结论:在临床常用腹腔镜气腹压力和作用时间范围内,CO2气腹激活相关信号转导通路,这可能是CO2气腹促肿瘤细胞生长机制之一。 Objective:To study the effects of CO2 pneumoperitoneum on the signal transduction pathways of human cervical carcinoma cell line CasKi. Methods:The imitated CO2 pneumoperitoneum model was established in vitro. After CasKi cells were exposed to CO2 pneumoperitoneum at 12,13 or 14mmHg for 2h,the cell growth and expressions of phosphor-ERK and phosphor-Akt were examined by MTl',immunocytochemistry and Western blotting respectively. The untreated CasKi cells served as control. Results:After treatment of CO2 pneumoperitoneum,the proliferation of human cervical carcinoma cell line CasKi significantly increased,and the expressions of phosphor-ERK and phosphor-Akt also significantly increased(P〈0.01 ). Among the CO: treated groups,the proliferation of human cervical carcinoma cell line CasKi and the expressions of phosphor-ERK and phosphor-Akt were not observed significant difference(P〉0.05). Conclusion:Under the pressure and during the exposure time which were often used in operational laparoscopy,CO2 pneumoperitoneum could activate correlated signal transduction pathways, which may be one reason for the promoting effect of CO2 pneumoperitoneum on malignant cells growth.
