摘要
目的:探讨胡芦巴种子水煎剂(胡芦巴)对实验性糖尿病大鼠肾脏基质金属蛋白酶-2(MMP-2)活性的影响。方法:利用链脲佐菌素(STZ)制作糖尿病大鼠模型,造模成功大鼠随机分为糖尿病模型组(DM)10只和胡芦巴组(FN)10只,另设10只正常大鼠对照组(N)。FN组糖尿病大鼠给予胡芦巴12周后,观察各组肾脏结构和MMP-2活性的变化。结果:光镜观察给予胡芦巴处理12周后,与DM组比较FN组肾小球病变减轻。免疫组化检测DM组肾小球ColⅣ表达明显增强,FN组ColⅣ表达明显低于DM组。FN组MMP-2活性形式酶解量为(1.41±0.18),与DM组(1.05±0.19)比较明显增多(P<0.05);与N组(1.53±0.25)比较差异无显著性。结论:胡芦巴可能通过增强肾脏MMP-2活性减轻实验性糖尿病大鼠肾脏病变的程度。
Objective To explore the effect of boiled fenugreek seeds on renal matrix metalloproteinase-2 (MMP-2) activity in diabetic rats. Methods The model of diabetes was built with STZ in rats. The model rats were randomly divided into diabetes control groups (DM) (n= 10) and fenugreek seeds groups (FN) (n= 10, and while normal control group (N) (n=10) rats was used. Diabetic rats were treated with fenugreek seeds for 12 weeks, the renal morphology and MMP-2 activity were observed in three groups . Results After diabetic rats were treated with fenugreek seeds for 12 weeks, optical microscopic examination indicated that the glomerular structure in N group was normal, the glomerular lesions in rats of DM groups were seriously and the pathologic changes of glomerular in rats of FN groups were alleviated significantly. Immunohistochemical results showed that the Col Ⅳ expression in glomerular ECM was increased in DM group compared with N group, and was decreased in FN group. The activity of MMP-2 was increased in FN group (1. 41±0.18) compared with DM group (1.05±0. 19) (P〈0. 05) and the alterations were not manifested compared with N group (1. 53±0.25). Cooclusion Fenugreek seeds may alleviate renal lesions through up-regulating MMP-2 activity.
