凋亡抑制蛋白Survivin在原发性肝细胞癌中的表达及其意义

Expression of Survivin in primary hepatocellular carcinoma and clinical significance

目的:通过研究凋亡抑制蛋白Survivin在原发性肝细胞癌(HCC)中的表达情况及其与临床病理特征之间的关系,探讨Survivin在HCC发生、发展中的作用。方法:采用流式细胞术(FCM)及免疫组化(SP)法对30例肝癌组织、20例癌旁组织及10例正常肝组织中的Survivin蛋白进行定量检测,并结合临床资料进行分析。结果:①Survivin在HCC组织中的表达率显著高于癌旁组织及正常肝组织(P<0.05);②Survivin蛋白在癌旁正常组织中无表达;③Survivin蛋白的表达与肿瘤的大小、部位、包膜、AFP无关联(P>0.05),而与镜下门静脉癌栓形成有关联(P<0.05)。结论:Survivin基因蛋白在HCC组织中过表达是反应HCC生物学行为的有效指标,Survivin基因蛋白表达的特异性有望成为HCC基因诊断与治疗的靶点。

Objective To investigate the expression of Survivin in primary hepatocellular carcinoma （HCC） and its relationship with clinical pathological features and explore the role of Survivin in the development of HCC. Methods The expressions of Survivin were detected in 30 cases of HCC, 20 cases of para-cancer tissue and 10 cases of normal liver tissue by flow cytometry （FCM） and immunohistochemistry （SP）. The clinical data were analyzed retrospectively. Results ① The expression of Survivin in the HCC tissue was significantly higher than those in para-cancer tissue and normal liver tissue （P〈0.05） ;②The expression of Survivin was negative in normal tissue surrounding cancer; ③The expression of Survivin was independence of tumor size, location, capsule, AFP （P〉 0.05）, while the expression of Survivin was related with endoscopic portal vein thrombosis formation （P〈0.05）. Conclusion The hyperexpression of Survivin in tissue can reflex the biological behavior of HCC. The specificness of survivin protein expression make it to be new target of tumor diagnosis and treatment.
