飞行员低密度脂蛋白受体基因NcoⅠ位点多态性对血脂的影响 被引量:2

摘要 目的了解飞行员低密度脂蛋白受体(LDL-R)基因NcoⅠ位点多态性及与血脂的关系,为飞行员高脂血症的防治提供依据。方法采用聚合酶链反应-限制性片断长度多态性方法,检测106例飞行员和98例健康对照者LDL-R基因NcoⅠ位点的多态性,并用常规方法测定其血脂水平。结果飞行员LDL-R基因NcoⅠ位点3种基因型-/-、-/+和+/+的频率分别为13.21%、48.11%和38.68%,-和+2种等位基因频率分别为0.373和0.627,与健康对照人群比较差异无显著性。飞行员的高脂血症发生率(52.83%)明显高于对照人群(29.59%,P<0.01)。飞行员-/-、-/+和+/+3种基因型的TC分别为(4.17±0.55)mmol/L、(4.59±0.61)mmol/L和(4.82±0.84)mmol/L,LDL-C分别为(2.18±0.48)mmol/L、(2.48±0.69)mmol/L和(2.74±0.75)mmol/L,其水平与基因型的关系为-/-、-/+向+/+递增,而TG和HDL-C在各基因型间无差异,与健康对照组相似。结论LDL-R基因NcoⅠ位点多态性是影响飞行员个体间血脂水平差异的独立遗传因素,但环境因素的作用也很重要。 Objective To investigate the relationship between low density lipoprotein receptor (LDL-R) gene Nco Ⅰ site polymorphism and blood lipid in the pilots, so as to provide the basis for preventing hyperlipidemia. Methods LDL-R gene Nco Ⅰ site genotypes were assayed by polymerase chain reaction-restricted fragments length polymorphism, and blood lipids were detected by routine methods in 106 pilots and 98 control subjects. Results The frequency of three genotypes ( -/- ,-/+ , and +/+ ) of LDL-R gene NcoI site in pilots was 13.21%, 48.11% ,and 38.68% respectively, - and + allele frequency was 0. 373, 0. 627 respectively. There was no significant difference in genotype and allele frequency of the pilots and control subjects. The incidence of hyperlipidemia (52.83 % ) of the pilots was much higher than that of the control (29.59%, P 〈 0.01 ). The levels of TC of three genotypes ( - / - , - / + , and +/+ ) in pilots were (4.17 ±0.55)mmol/L, (4.59 ±0.61)mmol/L, and (4.82±0.84) mmol/L respectively. The levels of LDL-C were (2.18 ± 0.48)mmol/L, (2.48 ± 0.69)mmol/L, and (2.74 ± 0.75) mmol/L respectively. The levels of TC and LDL-C were increased in the sequence of - / - , - / + , and + / + genotype. There was no difference in TG, and HDL-C among genotype groups. Conclusion LDL-R gene Nco I site polymorphism is an independent genetic factor on the pilot individual blood lipid levels, and environmental factors are important in inducing hyperlipidemia of the pilots.
