农杆菌介导遗传转化敏感基因型小麦的筛选鉴定 被引量:7

摘要 敏感型受体小麦品种鉴定的匮乏制约了农杆菌介导小麦的发展。以Gus基因检测及PCR检测为方法,对68个品种的成熟胚愈伤检测Gus基因表达,对8个品种的幼胚转化植株进行PCR检测,结果表明:受体材料扬麦10、师栾02-1、石家庄5号、周麦18、山东01-35、泰山008、Y9-63、邯郸3475经PCR检测,检测到目标基因,为农杆菌侵染敏感型受体材料;扬麦10、师栾02-1侵染后,Gus基因能够表达,且以恢复培养14 d的侵染效果最好,Gus基因表达的愈伤数量增加与蓝斑体积增大,可作为体系优化的受体材料。 The development of Agrobacterium-mediated transformation of wheat are restricted because the short of wheat genotype sensitive. This paper uses the method of the transformation expression of gus gene and PCR measure, detected 68 different materials of mature embryos in transformation expression of gus and 8 different materials of plants which were transformed from immature embryos, the results showed that foreign genes can be detect in Yangmail0, Shiluan02-1, ShijiangzhuangS, Zhoumail8, Shangdong01-35, Taishan008, Y9-63and Handan3475, so they are sensitive genotype materials and can be used for the receptor of the Agrobacterium infection; Gus genes were expressed in Yangmai10 and Shiluan02-1, the best effects of infection were resuming training in 14 d. The embryos members of Gus gene expression were increased, and the volume of locus caeruleus were augmented. So Yangmai10 and Shiluan02-1 can be used for the materials of the grading-up system.
