摘要
采用新型荧光试剂1,2-苯并-3,4-二氢咔唑-9-乙基氯甲酸酯(BCEOC)作为柱前衍生试剂,以甲醇乙腈为溶剂,醋酸铵、醋酸为电解液,首次建立了氨基酸衍生物的非水毛细管电泳分离新方法.该试验对甲醇、乙腈配比,醋酸铵、醋酸的浓度,柱温,电压进行了系统的研究,发现仅调整甲醇、乙腈的配比,改变醋酸铵、醋酸的浓度,都可显著的改善峰形、选择性和迁移时间等参数,使各组分得到有效的分离.在最优化条件下,实现了14种氨基酸衍生物的基线分离.建立的方法简单、快速.
A simple and rapid method for the separation of derivatized amino acids using 1,2-benzo-3,4- dihydrocarbazole-9-ethyl chloroformate(BCEOC)as derivatization reagent by non-aqueous capillary electrophoresis is proposed for the first time. To optimize the separation conditions, the background electrolyte composition is studied. With the mixed methanol and acetonitrile as the elution solvent, a satisfactory separation for the amino acid derivatives can be obtained when ammonium acetate and acetic acid are used as the electrolyte. At the same time, The effects of several key factors such as the proportion between MeOH and ACN, the concentrations of ammonium acetate and acetic acid on separation are also evaluated in this experimentation.
