摘要
目的探讨胰岛细胞移植后VCAM-1/VLA-4对移植物再血管化的影响。方法30只Wistar糖尿病模型大鼠分为糖尿病模型组、生理盐水组和免疫抑制剂组,其中糖尿病模型组常规喂食外未进行任何处理,生理盐水组和免疫抑制剂组进行了胰岛细胞移植,在术后分别采用生理盐水和免疫抑制剂进行处理。在术后每天固定时间进行外周血血糖测定,在术后10天和60天分别切取部分肝脏组织进行免疫组化分析肝脏组织中VCAM-1、VLA-4及F VIII-RA表达变化以观察三者间的关系。结果糖尿病模型组血糖变化不明显,术后NS组和免疫抑制剂组血糖均有下降,然而Ns组血糖逐渐上升接近糖尿病模型组,免疫抑制剂组血糖稳定;术后VCAM-1、VLA-4在NS组表达较糖尿病模型组及免疫抑制剂组增高,P〈0.05;术后FVIII-RA在免疫抑制剂组表达较糖尿病模型及NS组增高,P〈0.05;F VIII-RA同血糖相关性分析,A组:r=-0.097,P=0.684,无相关性:B组:r=-0.447,P=0.048,呈负相关性;C组:r=0.390,P=0.039,呈正相关性:VCAM-1/VLA-4与FVIII-RA相关性分析:各组间VCAM-1与FVIII-RA均无相关性,个组间VLA-4与FVIII-RA均呈正相关性。结论VCAM-1/VLA-4与胰岛细胞移植后的再血管化密切相关。
Objective To investigate the relationship between VCAM-1/VLA-4 and the graft revascularization after pancrea islet cell transplantation(PIT).Methods 30 diabetic Wistar male rats were divided into three groups equally at random, group A without any management but normal feeding: group B or group C were injected NS or CsA (3mg ·kg^-1·d^-1)+MMF(20mg · kg^-1·d^-1) intraperitoneally respectively after PIT. After PIT, plasma glucose in peripheral blood was detected by blood glucose meter: The expression of VCAM-1, VLA-4 and FⅧ-RA in hepatic tissue were monitored by immunohistochemistry 10 and 60 days after PIT to explore the relationgship between VCAM-1/VLA-4 and FⅧ-RA. Results There is no obviously changing of plasma glucose in peripheral around PIT in group A, plasma glucose in peripheral is stable in group B and C after PIT, but plasma glucose in peripheral increased gradually to the level of group A: The expression of VCAM-1 and VLA-4 in group A is higher than in group B and C in hepatic tissue after PIT: The expression of F VⅧ-RA in group C is higher than in group A and B in hepatic tissue after PIT: There is negative correlation between F Ⅷ-RA in hepatic tissue and plasma glucose in peripheral in groupB(r=-0.447, p=0.048), while positive
correlation between F Ⅷ-RA in hepatic tissue and plasma glucose in peripheral in group C (r=0.390, p=0.039); There is positive correlation between VLA-4 and F Ⅷ-RA in hepatic tissue in any group, while no correlation between VCAM-1 and F Ⅷ-RA in hepatic tissue in any group. Conclusion There is close correlation between VCAM-1/VLA-4 and revascularization after PIT.
