温度及冻融对壁虎抗H22肝癌作用的影响 被引量:5

摘要 目的:研究不同炮制温度及冻融对壁虎抗肝癌效果的影响,以明确较合适的壁虎炮制方法,指导临床应用。方法:MTT法检测37℃、75℃、100℃炮制的壁虎高、低剂量提取液对H22肝癌细胞体外杀伤作用。昆明小鼠H22移植瘤模型观察不同温度(37℃、75℃、100℃)炮制的壁虎高、低剂量组及非冻融壁虎组体内抑瘤活性及荷瘤小鼠胸腺、脾脏指数变化。结果:MTT法检测显示,壁虎不同温度高、低剂量组均可抑制H22细胞的增殖(P<0.05),高剂量组与西药抑制率差异无显著性意义(P>0.05)。体内实验显示75℃高剂量组抑制率45.8%与模型组比较差异有显著性意义(P<0.05)。37℃、100℃各组与模型组无统计学差异(P>0.05)。非冻融焙壁虎与干壁虎冻干粉抑瘤率分别为49.8%和51.1%,差异无显著性意义(P>0.05)。不同温度壁虎组,非冻融焙壁虎组,干壁虎冻干粉组与模型组小鼠的胸腺重、胸腺指数、脾脏重、脾脏指数相比均没有明显差异。而5-Fu组胸腺重、胸腺指数、脾脏重、脾脏指数较模型组和各壁虎组均明显降低(P<0.05)。结论:体内外实验均显示75℃恒温炮制能保持壁虎抗肿瘤活性成分,是有效的炮制温度。冻融并不是壁虎保持抗肝癌活性的必要条件。 Objective: To investigate the anti-tumor effects of different temperature processed gecko and none-freeze-thaw processed gecko on mouse H22 hepatocellular carcinoma to track down the appropriate method of gecko processing, Methods: MTT assay was applied to evaluate proliferation restraining effect of high and low dose of 37℃, 75℃, 100℃ temperature processed gecko on cultured H22 hepatoma cells in vitro. Using transplanted H22 tumor bearing mice model to observe the anti℃ tumor effect of different temperature processed gecko such as 37℃, 75℃, 100℃ and none-freeze-thaw gecko. Results: MTT assay showed both high and low dose of different temperature processed gecko revealed obvious anti-proliferation effect on the H22 cells (P 〈 0. 05 ) , There was no significant difference between high dose of different temperature processed gecko and 5-Fu group ( P 〉 0. 05 ) , In vivo experiment, the inhibition rate of high dose of 75 ℃ processed gecko was 45.8%, there was statistical differences compared with model gropu, However, there were no significant difference between 37℃, 100℃ processed gecko and model group (P 〉0. 05), the inhibition rate were 49, 8% and 51.1% respectively. There were no differences in thymus weight, thymus index, spleen werght, spleen index between among all gecko groups and the model group, However, the four observed items in 5-Fu groupwere all significantly lower than those in model group and all gecko groups (P 〈 0. 05 ) Conclusion: Experiments in vitro and in vivo showed that 75℃ processed gecko has good anti-cancer effect, freeze-thaw is not the essential requirement to maintain the anti-cancer effect of gecko.
