TPOAb液相竞争放免分析法的建立及临床应用

DEVELOPMENT OF A METHOD OF LIQUID COMPETITIVE RIA FOR TPOAb AND ITS CLINICAL USAGE

本文应用人甲状腺过氧化物酶(hTPO)单克隆抗体和hTPO单抗免疫亲和层析纯化的hTPO,用Iodogen法标记,建立了人甲状腺过氧物酶液相竞争的放射免疫测定。研究结果表明,本法灵敏度高,特异性强,重复性好,可取代MCA的测定,作为自身免疫性甲状腺疾病(AITD)尤其是桥本氏甲状腺炎诊断和疗效判断的重要指标。

Using human thyroid peroxidase (hTPO) monoclonal antibody (mAb) to purify hTPO through affinitive chromatography we labelled the hTPO with 125J by Iodogen and developed a method of liquid competitive RIA for TPOAb testing. The results showed that the method had high sensitivity, specificity and good reproducibility and it might replace MCA test for diagnosis and: clinical follow-up in autoimmune thyroid disease, especially Hashimoto thyroiditis.
