摘要
目的探讨结肠正常黏膜和腺癌的基因表达特征与差异。方法用基因表达系列分析(SAGE)技术构建两者的表达文库,分别挑选250和233个克隆进行测序,用SAGE2000软件进行分析和比较。结果构建了结肠正常黏膜和腺癌的SAGE文库,软件分析获得标签7926和7672个。在正常黏膜中高表达的36个基因主要与吸收、代谢、免疫等相关,在癌组织中有2个无表达,25个表达明显下降。在癌组织中高表达的38个基因主要是各种不同的核糖体蛋白,有32个表达升高。在癌组织中表达出现明显变化的还有多种核酸代谢及细胞周期调控基因。结论SAGE技术是构建表达文库的有效方法;结肠腺癌与正常黏膜高表达基因之间存在明显差异;在结肠癌中有多种与细胞周期调控、核酸代谢相关的基因表达发生改变。
Objective To explore the characteristics and difference of gene expression in adenocarcinoma and normal mucosa of colon. Methods The libraries of gene expression were generated by the technique of serial analysis of gene expression. 250 and 233 clones were selected and sequenced. The sequencing results were analyzed using the SAGE2000 software. Results 7926 and 7672 tags were gained from the sequencing results of SAGE libraries of normal mucosa and adenocarcinoma. Most of the 36 genes highly expressed in normal mucosa were genes related to absorption, metabolism and immunity. Among them,25 genes showed lower expression and 2 genes had no expression in cancer. Most of the 38 genes highly expressed in adenocareinoma were various kinds of ribosomal proteins and 32 of them had significantly higher expression than in normal mucosa. Many genes that participated in the metabolism of nudeonic acid and cell cycle control had significantly different expression between cancer and normal mucosa also. Conclusion The technique of SAGE is an effective method to generate the libraries of gene expression. The highly expressed genes in normal and cancerous tissues of colon were significantly different. The expression of many genes related to cell cycle control or metabolism of nucleonic acid changed in colon adenocarcinoma.
