三磷酸腺苷法肿瘤药敏检测与胃癌个体化化疗的临床研究

摘要 目的采用三磷酸腺苷生物荧光法(ATP-TCA)对胃癌组织标本进行肿瘤药敏检测,为胃癌的个体化化疗选择提供依据。方法42例胃癌新鲜标本行肿瘤细胞分离,原代培养后用ATP-TCA技术对5种6组化疗药物进行检测。结果所有标本均顺利完成检测,可评估率为100%。不同胃癌患者对同一种化疗药物的敏感度显著不同,差异有高度统计学意义(P<0.01);不同胃癌患者对不同药物的敏感程度也不同,差异亦有高度统计学意义(P<0.01)。结论ATP-TCA可为胃癌的临床个体化化疗提供重要的指导作用。 Objective To investigate the heterogeneity of chemosensitivity in gastric cancer using adenosine triphosphate based tumour ehemosensitivity assay(ATP-TCA) and the feasibility of individual chemotherapy. Methods ATP-TCA was used to determine the effect of 5 single drugs and 20 times 5-Fu added with L-OHP imitated Xeloda in surgical specimens from 42 patients with gastric cancer. Results All above these 42 specimens were complete detected successfully and the rate of evaluation was 100%. There was considerable difference in ehemosensitivity between individuals. There was the heterogeneity of in vitro ehemosensitivity in gastric cancer and had marked statistical significance (P〈0.01). Conclusion The use of the ATP-TCA may provide a method of selecting appropriate anti-cancer drugs in gastric cancer.
