乳腺浸润性导管癌中COX-2、MMP-2的表达与细胞增殖的关系

Expression of COX-2 and MMP-2 in Invasive Ductal Carcinoma of Breast and Its Relationship with Cell Proliferation

目的探讨环氧化酶-2(COX-2)和基质金属蛋白酶-2(MMP-2)蛋白在乳腺浸润性导管癌组织中的表达及其与细胞增殖的关系。方法应用免疫组织化学法检测52例乳腺浸润性导管癌组织中COX-2、MMP-2和Ki-67蛋白的表达情况。结果乳腺浸润性导管癌组织中COX-2的阳性表达率为57.7%(30/52),MMP-2的阳性表达率为82.7%(43/52),在乳腺浸润性导管癌组织中COX-2、MMP-2蛋白的表达之间存在显著正相关(r=0.498,P<0.01)。COX-2、MMP-2阳性组Ki-67指数明显高于COX-2、MMP-2阴性组,差异有统计学意义(P<0.05)。结论乳腺浸润性导管癌组织中存在COX-2、MMP-2的高表达。COX-2蛋白可通过诱导MMP-2蛋白的表达上调,增加乳腺癌细胞的侵袭力,并促进肿瘤细胞增殖,从而成为其促进乳腺癌浸润、转移的因素之一。

Objective To study the expression of cyclooxygenase-2 （COX-2） and matrix metallproteinase-2（MMP-2） in invasive ductal carcinoma of breast and its relationship with cell proliferation. Methods Expression of COX-2, MMP-2 and Ki-67 was detected in 52 cases of breast cancer tissue by immunohistochemistry. Results The positive COX-2 expression was 57.7% in invasive ductal carcinoma of breast, and the positive MMP-2 was 82.7%. There was a significant correlation between the expressions of COX-2 and MMP-2 in breast cancer （r=0.498,P〈0.01）. In the response group of COX-2 and MMP：2 protein Ki-67 index was higher than that in non-response group（P〈0.05）. Conclusion The overexpression of COX-2 and MMP-2 protein is found in invasive ductal carcinoma of breast.To stimulate MMPs especially MMP-2 overexpression may be one of the pathways of COX-2 protein to contribute to invasion and metastasis of human breast cancer.
