摘要 基于均匀设计实验,对重楼总皂苷的提取工艺进行了优选.以重楼总皂苷含量为指标,选用U8(85)为均匀设计实验方案,考察了加热回流时间、提取液乙醇浓度、浸泡时间及提取次数对重楼总皂苷提取的影响.采用AB-8型大孔吸附树脂对粗皂苷进行吸附、除杂和洗脱,收集洗脱液,并用HPLC对洗脱液进行含量测定.实验结果表明最佳的工艺条件为:2g滇重楼粗粉(40目)用40%的乙醇浸泡30小时后,加热回流20m in,抽滤,药渣再重复加热回流7次,合并滤液,减压蒸馏,得粗皂苷浓缩液. The extraction process of steroidal saponin from Paris polyphylla has been investigated using the uniform design of U8 ( 8^5 ). With the content of steroidal saponin as the detecting index, extraction time, soaking time, ethanol concentration and times of extraction the four factors have been optimized. AB-8 macroporous adsorptive resin was used to adsorb steroidal saponin, remove the impurity and elute steroidal saponin. The eluted solution of steroidal saponin is analyzed by using HPLC. The experiments show that the optimum condition for the extraction of 2g Paris polyphylla powder is : 30 hours of soaking time, 40% of ethanol concentration, 20 min of extraction time, and 8 times of extraction.
