枸杞多糖对2型糖尿病胰岛素抵抗模型大鼠resistin基因表达的影响 被引量:8

摘要 观察枸杞多糖(LBP)对高脂膳食加链脲佐菌素(STZ)诱导的2型糖尿病(2-DM)大鼠胰岛素抵抗指数及肾周脂肪组织抵抗素(resistin)基因表达的影响。选用高脂饲料加STZ建立2-DM胰岛素抵抗大鼠模型,将造模大鼠随机分为模型对照组、罗格列酮组和LBP高、中、低剂量组(80mg/kg、40mg/kg、20mg/kg),每组各10只,另选10只大鼠为正常对照组。各组灌胃8周后,空腹取材,RT-PCR法检测肾周脂肪组织resistin基因mRNA水平,放射免疫法测定空腹胰岛素并计算胰岛素抵抗指数(insulin resistance index,IRI)。结果表明,与模型组比较,罗格列酮组及LBP各剂量组均能显著降低2-DM大鼠肾周脂肪组织resistin基因mRNA表达,降低IRI。说明LBP具有改善2-DM大鼠胰岛素抵抗的作用。 To explore the effect of lyciurn barbarurn polysaccharide on mRNA expression of resistin gene in perirenal adipose tissue and insulin resistance index of 2-DM rats induced by high-fat diet and strep tozotocin (STZ). The model of type 2-DM IR rats induced by high-fat diet and STZ was built. The 2-DM model rats were divided into five groups at random, model group, rosiglitazone group and high, middle, low doses of LBP groups with 10 rats in each group. Another 10 rats were selected as control group. Serum insulin was detected by radioimmunoassay and insulin resistance index (IRI)was calculated, and mRNA expression of resistin gene in perirenal adipose tissue was determinded by RT-PCR after rats were fed for eight weeks. The results showed that compared with the model group, the mRNA expression of resistin gene was lower in the rosiglitazone group and three doses of LBP groups(P 〈 0. 01), and IRI was also decreased (P 〈0. 01). It demonstrates that LBP can improve insulin resistance of 2-DM rats.
