摘要
研究大头金蝇蛆提取物对白血病和肺癌细胞的抑制作用。采用MTT法对大头金蝇蛆干粉提取物不饱和脂肪酸、粗脂肪和蝇蛆干粉分别作体外抑瘤试验,观察到了不饱和脂肪酸、粗脂肪、和蝇蛆干粉分别在剂量125μg/ml、250μg/ml和500μg/ml时对人原髓细胞白血病HL-60有明显的增殖抑制作用,其中不饱和脂肪酸剂量250μg/mL时其增殖抑制率为96.2±3.6%;该剂量对人肺腺癌A549细胞的增殖抑制率为97.0±0.5%,而蛋白质组份对上述两种肿瘤细胞无增殖抑制作用。表明大头金蝇蛆提取物对该两种癌细胞的增殖抑制作用其有效组份是脂肪酸,尤其是不饱和脂肪酸。不饱和脂肪酸急性毒性试验LD50>20g/kg(为人服用量的1000倍以上),为寻找抗白血病和肺癌的安全有效物质提供了一定实验依据。
To study the inhibition of Chrysomyia Megacephala Fab larva extracts on lung cancer cell A549 and Leukemia cell HL-60. The effect of the unsaturated fatty acid, rouht fat, larva dry powder and the extracts of Chrysornyia Megacephala Fab larva on lung cancer cell A549 and Leukemia cell HL-60 were observed by cell culture in vitro using the method of MTT. When culturing HL-60 and the extracts with different concentrations, they had obvious inhibition action on cell activity with concentrations of 125,250,500 μg/ml. Data showed that the Inh% of unsaturated fatty acid was (96.2±3. 6)% with a concentration of 250 μg/ml. With the same concentration, the Inh% of unsaturated fatty acid on A549 was (97.0±0.5)%. However the protein was not detected. This showed that effective component of Chrysomyia Megacephala Fab larva was fat, especially unsaturated fatty acid. LD50 of the unsaturated fatty acid was above 20 g/kg(1000-fold more than human daily dose ),which afforded experiment information for low-oxic and efficient drugs of antileukemia and lung cancer.
