丙三醇缩水甘油醚交联脱细胞猪真皮基质的条件优化 被引量:2

Study on optimized condition of porcine acellular dermal matrix cross-linked by glycerol polyglycidyl ether

摘要 运用正交设计法研究了丙三醇缩水甘油醚(glycerol polyglycidyl ether,GPE)交联脱细胞猪真皮基质材料(porcine acellular dermal matrix,pADM)的基本性能.考察了温度、pH值、用量和反应时间对改性材料(GPE-pADM)收缩温度的影响,并对优化条件下改性后的材料进行了性能表征.实验结果表明:温度为37℃,pH值10.5,反应时间72 h,GPE用量20%,催化剂为三乙醇胺改性条件为最佳.GPE交联后材料的收缩温度明显升高,自由氨基含量降低,抗张强度和断裂伸长率降低,接触角显著降低,亲水性明显增强.扫描电镜结果表明,交联对材料孔隙结构未产生明显影响. The basic performance of porcine acellular dermal matrix(pADM)were studied after cross-linked by glycerol polyglycidyl ether with the method of orthogonal experiment. The relation between reaction conditions and shrinkage temperature(Ts) of cross-linked material were discussed preliminarily, through researching on Ts of the GPE-pADM influenced by the different temperature, pH, dosage and reaction time. In order to prove the result, the performance of GPE-pADM were mensurated, such as Ts, content of free amino, tensile strength and elongation at break, contact angle and SEM. It was showed that the comprehensive performance was best when the reaction temperature was 37 ℃, the pH was 10.5, the reaction time was 72 h, the catalyzer was triethanolamine (TEA) and dosage was 20%. It was showed that Ts of GPE-pADM was raised obviously, the content of free amino, tensile strength and elongation at break were declined. The hydrophilic performance of GPE-pADM had been improved after cross-linking which was approved by the reducing of contact angle. The porous framework of pADM almost was not changed after cross-linked.
