摘要
目的尼莫同对研究颅脑损伤后AQP4表达和血脑屏障(BBB)通透性的影响。方法健康成年Wistar大鼠50只,随机分成尼莫同组和生理盐水对照组。自由落体硬膜外撞击方法致重度脑创伤模型。在伤后6h、12h、24h、48h和72h每组各处死5只动物。取大鼠受损脑组织用于以下实验:①HE染色进行形态学观察;②免疫组化检测AQP4的表达变化;③测伤区脑组织中伊文氏蓝(EB)外渗的量,以反映伤侧BBB通透性的变化。结果颅脑损伤后,在尼莫同组和对照组损伤的脑组织均出现脑损伤的病理改变、AQP4表达上调、BBB通透性增加等改变,其变化趋势一致。颅脑损伤后6h、12h、24h、48h和72h,尼莫同组损伤脑组织EB外渗量明显多于对照组(P<0.05)。结论尼莫同可以增加颅脑损伤后BBB通透性,加重脑损伤,但不影响受损脑组织AQP4的表达。
Objective To investigate the effect of nimotop on the expression of aquaporin--4 (AQP4) and the permeability of blood-brain barrier(BBB) after traumatic brain injury. Methods Fifty healthy Wistar rats in whom traumatic brain injury was made by Feeney et al method were randomly divided into Nimotop (NIM) group and the control group. Twenty-five of each group were divided into 5 subgroups, i.e. 6, 12, 24 hours, 48 hours, 72 hours subgroups after TBI. The morphological changes in the injured cerebral tissues were observed under a light microscope. The expression of AQP4 was determined by immunchitohemical technique. The BBB permeability of the traumatic brain tissue was determined by Evans blue(EB) extravasation method. Results The morphological changes in the cerebral tissues and the expression of AQP4 in NIM group were similar to that in the control group after TBI. The contents of EB in the injured cerebral tissues were significantly higher in NIM group than those in the control group 6, 12, 24, 48, 72 hours after TBI (P〈0.05). Condusions NIM can increase the permeability of BBB after TBI. However, NIM has no effect on the expression of AQP4 in the injured cerebral tissues after TBI.
