摘要
目的研究白血病细胞株HL-60中基质金属蛋白酶2、9(MMP-2、MMP-9)和基质金属蛋白酶天然抑制剂-1(TIMP-1)的表达及细胞因子干扰素(IFN-γ)对MMP-2、MMP-9和TIMP-1转录水平的调节。方法分别在HL-60细胞生长的不同时期(1-5d)收集细胞,采用半定量RT-PCR方法,分析白血病细胞株HL-60中MMP-2、MMP-9和TIMP-1mRNA的表达水平;以不同浓度的IFN-γ作用于HL-60细胞,24、48h后RT-PCR检测MMP-2、MMP-9和TIMP-1mRNA水平的变化。结果细胞接种后24hMMP-2和MMP-9的mRNA水平最高,但TIMP-1表达相对恒定。IFN-γ各个浓度组对MMP-2和MMP-9mRNA表达均有抑制作用(P<0.05),且随着作用浓度的增高mRNA表达减少;TIMP-1对其调节不敏感。结论明确了白血病HL-60细胞株在转录水平表达MMP-2、MMP-9和TIMP-1;IFN-γ对MMP-2、MMP-9mRNA表达有明显的抑制作用,而对TIMP-1转录无明显作用。
Objective To detect the expression of MMP-2, MMP-9 and TIMP-1 in leukemia cell line HL-60, and explore the effect of interferon- γ (IFN- γ ) on their expression. Methods Reverse transcriptase-polymerase chain reaction was used to assay MMP-2, 9 and TIMP-1 mRNA in leukemia cell line HL-60 at different time (the first day to the fifth day of the cell growth), and half-quantity values of the influence on the cells after treatment with IFN- γ in different concentration and different time (24 hours, 48 hours). Results The expression of MMP- 2, -9 mRNA was significantly the highest after 24 hours of the cell culture. But the expression of TIMP-1 mRNA was constant. IFN-γ in different concentration could down-regulate the mRNA expression of MMP-2, -9 (P〈0. 05), and as the concentration increased, mRNA expression decreased. But mRNA expression of TIMP could not be affected by IFN-γ. Conclusion MMP-2, -9 and TIMP-1 mRNA are expressed in leukemia cell line HL-60. IFN- γ can inhibit expression of MMP-2 or MMP-9, but has no influence on TIMP-1 mRNA expression.
