摘要
目的建立人卵巢癌裸鼠移植实体瘤模型。方法将1例人卵巢癌组织移植裸鼠,建立人卵巢癌裸鼠原代移植实体瘤模型的基础上,再将实体瘤皮下移植、实体瘤原位移植、实体瘤细胞移植裸鼠。观察裸鼠实体瘤生长和转移情况,称量其体重、子宫卵巢重、瘤重及瘤的大小,并作病理、电镜、染色体检查。结果成功地建立人卵巢癌裸鼠移植实体瘤模型,并已传至第18代,传代移植成功率100%,组织学和超微结构形态均证明该实体瘤保留了原人卵巢癌特征,有人卵巢癌染色体特征,并出现肝、脾转移。结论本研究建立人卵巢癌裸鼠移植实体瘤模型与人相似,通过18代传代和实验观察方法稳定可靠。为人卵巢癌的研究提供了理想的动物模型。
Objective To establish human ovarian cancer transplanted subcutaneously nude mice model. To establish a nude mouse model of human ovarian cancer by subcutaneous transplantation. Methods Tissue samples from a human ovarian cancer were transplanted into nude mice, from which the tumor was further transplanted into other nude mice by subcutaneous or orthotopic transplantation for many times. The tumor growth and metastasis in the nude mice were observed, the body weight, the weight of ovaries and tumors and tumor size were measured, the tumor cell karyotype was analyzed, and the tumor tissues were examined by histopathology and transmission electron microscopy. Results A nude mouse model of human ovarian cancer was successfully established by subcutaneous transplantation. This model was already passaged for 18 generations with a 100% success rate of transplantation. Histological and uhrastructural examination revealed that the model remained human ovarian cancer characteristics, such as human ovarian cancer chromosome karyotype, and occurrence of liver and spleen metastases. Conclusion A nude mouse model of human ovarian cancer has been successfully established. The high success rate of 18 passages and the biological characteristics of the tumor model demonstrate that it may provide a stable, reliable and useful animal model in human ovarian cencer research.
