p57^(KiP2)和Cyclin A在喉癌中的表达及其临床意义 被引量:3

摘要 目的研究细胞周期素依赖性激酶抑制因子p57KiP2和细胞周期素A(CyclinA)在喉癌组织中的表达及其临床意义。方法40例喉癌、20例声带不典型增生、20例正常喉组织切缘标本,免疫组化S-P法检测p57KiP2和CyclinA的表达活性,比较分析其临床意义。结果在喉癌组织中,p57KiP2阳性表达率42.5%,显著低于癌旁正常喉黏膜与不典型增生喉黏膜(P<0.05),并与喉癌组织的分化程度和淋巴结转移特性相关(P<0.05);CyclinA阳性表达率55.0%,明显高于癌旁正常喉黏膜与不典型增生喉黏膜(P<0.05),并与喉癌组织的分化程度和淋巴结转移特性相关(P<0.05)。p57KiP2与CyclinA的表达活性呈负相关(P<0.01)。结论p57KiP2的低表达及CyclinA高表达在喉癌的发生、发展过程中可能发挥重要作用,其表达水平与喉癌的分化程度、淋巴结转移与否及预后密切相关,是判断喉癌恶性程度与淋巴结转移特性的重要参考指标。 Objective To investigate the clinical implications of expressive activities of p57^KiP2 and cyclin A in the tissue samples of laryngeal cancer based on a study carried out in a group of cases with the lesion. Methods Included in this study were 40 samples of laryngeal cancer, 20 ones of vocal cords with atypical hyperplasia and 20 ones of healthy laryngeal mucosa at the cutting edge following the focus of cancerous neoplasm removed. Then, immunohistochemical staining SP procedures were performed in these samples to determine the expressive activities of p57^KiP2 and cyclin A, followed by a comparative analysis on their expressive differences and clinical implications. Results In the tissue samples taken from laryngeal cancer, the positive rate of p57^KiP2 was 42.5%, significantly lower than that in the healthy laryngeal mucosa near to laryngeal cancer and that in vocal cords with atypical hyperplasia (P〈0.05), with a marked correlation with the differentiating degree of laryngeal cancer cells and the metastatic tendency of the lesion to neck nodes. In contrast, the positive rate of cyclin A was 55.0% in the cancerous tissues,significantly higher than that in both of the latter two kinds of tissue samples (P〈0.05), with a marked correlation with the differentiating degree of laryngeal cancer cells and the metastatic tendency of the lesion to neck nodes as well. On the other hand, there was a negative correlation of the expressive activity of p57^KiP2 with that of cyclin A in the lesion tissues themselves. Conclusions The down-regulated expressive activity of p57^KiP2 and the up-regulated one of cyclin A may both play their important role in the initiation and development of laryngeal cancer, with their expressive levels more closely correlated with the differentiating degree of cancer cells and the metastatic tendency to neck nodes. Therefore, they can be taken as the important reference markers for evaluating the malignant level and the metastatic tendency of this kind of tumor as well.
