摘要
利用少根根零发酵生产富马酸,考察了不同浓度的初始葡萄糖以及有机氟源酵母膏和无机氮源(NH_4)_2SO_4对富马酸的合成的影响。在此基础上运用响应曲面分析方法优化了富马酸产量以及转化率,葡萄糖和(NH_4)_2SO_4的浓度为162.0g/L、3.08g/L时,富马酸的产量最大为75.15g/L;葡萄糖和(NH_4)_2SO_4的浓度为121.2g/L、2.23g/L时,富马酸的转化率最大为55.16%,在原来基础上产量有了明显的提高,优化效果较好,通过验证实验值与预测值基本相符。
The production and yield of fumaric acid by Rhizopus arrhizus cultivated on different concentrations of glucose as carbon source and (NH4)2SO4 and yeast extract as nitrogen sources were investigated. As the concentration of initial glucose and (NH4)2SO4 has important influence on biosynthesis of fumaric acid. They were optimized in shake flasks using response surface analysis to determine the optimal levels for production and yield. The maximum fumaric acid production (75.15 g/L) was obtained consistently in flask fermentation using the optimized concentrations of glucose and (NH4)2SO4 (162.0 g/L and 3.08 g/L, respectively), and the maximum yield (55.16%) was obtained using the optimized concentrations of glucose and (NH4)2SO4(121.2 g/L and 2.23 g/L, respectively).
