嗜热蛋白酶生产菌DPE7的16 S rRNA基因克隆及系统发育

摘要 [目的]确定嗜热蛋白酶生产菌DPE7的系统发育地位。[方法]通过PCR方法扩增出嗜热蛋白酶生产菌DPE7的16 S rRNA基因片段,并对其进行了克隆和测序。[结果]对该序列在GenBank中的BLAST结果表明,相似性高于99%的序列中大部分是泥土芽胞杆菌的16 S rRNA基因序列,其中与Geobacillus toebii T1680的16 S rRNA基因序列相似性达99.60%。对菌株DPE7和其他13株泥土芽胞杆菌的16 S rRNA基因序列进行系统发育分析,菌株DPE7与其中的4株泥土芽胞杆菌聚类在一起。[结论]经16 S rRNA基因序列同源性比较和系统发育分析,确定菌株DPE7为泥土芽胞杆菌。 [ Objective] The research aimed to ascertain the phylogenetic position of strain DPET. [ Method] The 16 S rRNA gene fragment of strain DPET, was amplified with PCR, cloned and sequenced. [Result] The BLAST of 16 S rRNA gene sequence in GenBank indicated that the species with close similarity to strain DPE7 were Geobacillus and the similarity value of 16 S rRNA gene sequence between strain DPE7 and Geobacillus toebii T1680 was 99.60%. The phylogenetic tree was produced based on the 16 S rRNA gene sequence of strain DPE7 and 13 Geobacllus species. It showed that the strain DPE7 was clustered with 4 Geobacillus species. [ Conclusion] The swain DPE7 should belong to Geobacillus by homology comparison and phylogenefic analysis.
