羟基喜树碱固体自微乳和速释微球结肠定位胶囊的大鼠体内药动学 被引量:2

摘要 目的:研究羟基喜树碱(HCPT)固体自微乳和速释微球的结肠定位胶囊在大鼠灌胃给药后的药动学特征和结肠组织的药物浓度,以提高靶位药浓,降低全身毒副作用。方法:分别制备羟基喜树碱固体自微乳(SSMEDDS)及速释微球(microsphere),将它们和HCPT原药分别灌装于大鼠结肠定位胶囊中;分别以6 mg.kg-1的剂量对大鼠灌胃给药,后于不同时间点进行眼眶静脉丛取血,用HPLC法测定大鼠体内HCPT含量,并用3P97药动学程序对血药浓度进行处理;最后测定给药大鼠结肠组织的血药浓度。结果:HCPT对照液和结肠给药的HCPT、HCPT-SSMEDDS、HCPT-microsphere的AUC分别为(329.8±5.6)ng.h.mL-1、(134.9±3.4)ng.h.mL-1、(221.3±6.9)ng.h.mL-1和(207.6±4.8)ng.h.mL-1;MRT分别为(2.9±0.2)h、(4.8±0.4)h、(6.2±1.4)h和(5.7±1.2)h;结肠组织中内酯浓度分别为:(1.7±0.2)μg.g-1、(4.3±0.4)μg.g-1、(13.7±1.4)μg.g-1和(8.9±0.7)μg.g-1。结论:HCPTK-SSMEDDS结肠给药后可以显著降低大鼠全身血药浓度,提高结肠组织的血药浓度,并且相对于HCPT-microsphere,内酯型HCPT的稳定性提高,为HCPT治疗结肠癌提供了新的选择。 Objective: To study pharmacokinetic characteristics of colon specific capsule of HCPT (hydroxycamptothetin)-SSMEDDS( solid self-emulsifying drug delivery systems) and HCPT-microsphere in rats. Methods: HCPT-SSMEDDS, HCPT-microsphere and raw HCPT were filled into colon specific capsule respectively. Plasma concentrations of HCPT in there forms were detected at various times following a single ig close of 6 mg/kg HCPT. The concentration of 10-HCPT in plasma was determined by HPLC. Experimental data were processed by the pharmacokinetic software 3P97. Results: Pharmacokinetic parameters of HCPT control, colon specific capsule of HCPT, HCPT-SSMEDDS, HCPT-microsphere were as follows: AUC (329.8±5.6) ng.h.mL^-1, (134.9±3.4) ng.h.mL^-1, (221.3±6.9) ng.h.mL^-1 and (207.6± 4.8) ng.h.mL^-1 respectively; MRT (2.9±0.2)h, (4.8 ±0.4) h, (6.2±1.4) h and (5.7 ±1.2) h respectively; concentration in colon tissue (Clactone ) : (1.7 ±0.2) μg.g^-1, (4.3 ±0.4) μg.g^-1 , (13.7 ± 1.4) μg.g^-1 and (8.9 ± 0. 7 ) μg.g^-1 respectively. Conclusion: Colon specific capsule of HCPT-SS- MEDDS can decrease the Clactone in plasma, and increase Clactone in colon tissue.
