产酶溶杆菌OH11菌株摇瓶发酵条件研究 被引量:5

摘要 采用单因素试验法研究了温度和转速等因子对产酶溶杆菌OH11液体发酵的影响。结果表明温度以30℃为宜,转速以210r/min为宜。通过正交试验在摇瓶中以LB为基础培养基对发酵培养基进行了优化。优化后的培养基中活菌数在48h时可达到4.5×109cfu/ml,明显高于原基础培养基的结果;通过正交试验对其他发酵条件的优化结果表明,装液量40ml/250ml、发酵时间72h、接种量108cfu/ml和初始pH值7.5为最适条件。 The fermentation of Lysobacter enzymogenes OH11 was investigated through single-factor test. The main factors include: temperature, rotate speed. The optimal fermentation conditions are: temperature 30℃, rotate speed 150 r/min. The orthogonal experiment was used to optimize the medium, and the microbe number of OH11 could reach 4.5 × 10^9 cfu/ml under optimal condition after 48 h. The fermentation condition of OH11, such as volume, fermentation time, inoculum' s size and pH value before sterilization were tested by orthogonal experiment. The optimum fermentation conditions are as follows: volume 40 ml/250 ml, fermentation time 72 h, inoculum size 10^8 cfu/ml, pHT.5 before sterilized.
