摘要
目的建立蝉花菌丝体中多球壳菌素含量的固相萃取结合高效液相色谱测定法。方法高效液相色谱法,甲醇超声提取,固相萃取小柱纯化,ODS柱,流动相为乙腈-水(65∶35),流速1.0mL·min-1,柱温30℃,检测波长203nm。结果多球壳菌素分离良好,加样回收率为98.95%,RSD为2.22%(n=6),在0.15~7.54μg内呈现良好的线性关系。结论该方法简便易行、灵敏、重现性好,可用于蝉花菌丝体中多球壳菌素含量的测定。
OBJECTIVE To establish a sensitive method for the determination of the content of myriocin in samples collected from submerged cultivation in C. cicadae. METHODS Sample was extracted with methanol and solid phase extraction, and identified by MS. Myriocin in C. cicadae was separated on a ODS column. The mobile phase was acetonitrile-water (65:35 ) at a flow rate of 1.0 mL· min^-1. The UV detection wavelength was 203 nm. RESULTS Myriocin in sample solution was well separated. The average recovery was 98, 95 % with RSD 2. 22% (n = 6) and the linear range was 0. 15 -7.54 μg. CONCLUSION The method is convenient, rapid, accurate, and sensitive, thereby leading to a wide and effective analysis for the determination of the myriocin-like immunoregulational ingredient in C. cicadae.
