摘要 基质效应是指在 LC/MS 测定中,色谱分离时共洗脱的物质改变了待测成分的离子化效率,所引起的信号的抑制或提高。由于生物样品中存在大量内源性干扰物质,基质效应的影响更为显著。本文对生物样品 LC/MS 分析测定中基质效应产生的机制、测定基质效应的方法、减小基质效应的措施以及在方法学验证中如何考察基质效应进行了综述。 Matrix effects are the alternation of ionization efficiency by the presence of coeluting substances. These effects in the chromatogram are more significant in biological matrices and have deleterious impact on method accuracy and sensitivity. The exact mechanism is still unknown, the possible sources of matrix effects and the methods to remove or minimize it in method development are discussed in this paper. In addition, the assessments of matrix effects or relative matrix effects in method validation are also summarized.
