摘要 目的:探讨高脂血症对急性胰腺炎(AP)的影响及其可能的机制。方法:分别用triton WR1339、caerulein制作大鼠高脂血症和AP模型,同时应用两者制作伴有高脂血症的AP模型,比较各组胰腺病理损害评分、腹水量、胰腺湿/干比、血清淀粉酶和胰腺组织的凋亡;Western blot检测胰腺组织蛋白激酶C(PKC)的膜转位。结果:Triton WR1339诱导大鼠高脂血症以甘油三酯升高为主,6h达蜂,升高达20倍,使部分大鼠出现轻型AP。T+AP组病理评分、腹水量、胰腺湿/干比和血清淀粉酶较AP组均显著升高(P<0.05)。TUNEL法测得伴有高脂血症的AP腺泡出现凋亡数量最多。T+AP组PKC膜转位比例最高。结论:高脂血症可以诱导AP或加重AP的胰腺损伤。PKC的活化可能是高脂血症加重AP的机制之一。 Objective To investigate the effects of hyperlipaemia on acute pancreatitis (AP) and the possible mechanisms. Methods Hyperlipaemia and AP (AP group) models were introduced by triton WR1339 and caerulein respectively. AP with hyperlipaemia model (T + AP group) was prepared by both triton WR1339 and caerulein simuhaneously. In each group, we compared the histological score, volume of ascites, ratio of pancreatic wet/dry weigh, serum amylase and pancreatic acinar cell apoptosis. The level of protein kinase C (PKC) membrane translocation in pancreatic tissue was detected by Western blot, Results In the hyperlipaemia model introduced by triton WR1339, triglyceride was increasing remarkably and reached apex about 20 times than control group at 6 hours after injection, and most rats had mild acute pancreatitis. Histological score, volume of ascites, ratio of wet/dry weigh and serum amylase in T + AP group were obviously higher than AP group (P 〈 0.05). Apoptosis cells detected by TUNEL increased in T + AP group. PKC membrane translocation level was enhanced mostly in T + AP group. Conclusions Hyperlipaemia may induce AP or intensify pancreatic injury of AP. PKC activation may be one of the mechanisms of AP intensified by hyperlipaemia.
