摘要
目的研究大鼠骨髓间充质干细胞(rat mesenchymal stem cells,rMSCs)在体外诱导为神经源性细胞的可行性,并探讨其作用机制。方法从Wistar大鼠骨髓中获得rMSCs,纯化培养传代后用不同浓度的bFGF诱导,经MTT法检测bFGF对rMSCs生长的影响;用倒置光显微镜、透射电镜、免疫组化和RT-PCR等方法鉴定诱导后细胞行为改变与snail mRNA表达。结果bFGF对rMSCs具有促增殖作用,在倒置光显微镜和透射电镜下可见到有神经胶质样细胞形成,免疫组化证明GFAP的表达较对照组显著增高(P<0.01)。RT-PCR结果表明,诱导组细胞snail mRNA明显表达。结论bFGF促进rMSCs转分化为GFAP阳性的神经胶质样细胞,转录因子Snail可能与转化过程有关。
Objective To investigate the feasibility of inducing rat measenchymal stem cells (rMSCs) into nerve tissue cells in vitro and its mechanism. Method The rMSCs were taken out from rat bone marrow and cultured. The passaged cells were exposed to different concentrations of basic fibroblast growth factor (bFGF) . The cell proliferation was examined by methyl-tetrazolium(MTT) assay. The behavior changes of rMSCs were detected by light microscope, transmission electron microscope and immunocytochemistry. The Snail mRNA expression was detected by RT-PCR. Result bFGF improved the proliferation of rMSCs significantly, the neuroglia-like cells were seen by light microscope and transmission electron microscope in bFGF group. The percentages of GFAP-positive cells were increased significantly in bFGF group than in control(P〈 0.01). The expression of Snail mRNA was found obviously in transdifferentiated cells by bFGF but weaker in control. Conclusion bFGF can promote the proliferation and differentiation of MSCs into glial phenotypes in vitro ,Snail may be involved in the process.
