昆明小鼠四倍体胚胎的制作

摘要 目的介绍一种简单易行的制作四倍体胚胎的方法。方法取腹腔注射人绒毛膜促性腺激素(hCG)后37~40h的小鼠2-细胞胚胎,应用电融合方法,制作四倍体胚胎。用CZB培养液对四倍体胚胎进行体外培养。结果采用交流电((AC)3V/mm、直流电(DC)140V/mm、脉冲宽度50s、一次脉冲进行融合,融合率为97%,囊胚率为72.7%。结论电融合方法可以应用于制作四倍体胚胎。 Objective To introduce a simple tetraploid embryo produced method. Method Mouse 2-cell embryo was obtained after peritoneal injection of human chorionic gonadotropin (hCG) for 37- 40 hours, mice tetraploid embryo was produced by electrofusion. Result Tetraploid embryo was electrofused successfully with alternating current(AC)3V/mm, direct current (DC) 140V/mm, pulse duration 50μs and one time pulse, the rate of fusion was 97%, the rate of blastocyst was 72.7%. Conclusion Electrofusion method can be applied to produce tetraploid embryo.
