骨髓微环境造就的干细胞凝集态骨髓细胞球

摘要 细胞分化受到细胞自发信号和周围环境的调控。体内的细胞与细胞、细胞与细胞外基质形成了复杂的微环境,而微环境对于骨髓中的干细胞和前体细胞的存活、分化、增殖和迁移都起着重要的作用。成人的造血过程发生在骨髓腔中,造血细胞与其周围环境始终呈动态变化过程。在骨髓腔中的稳态下,大部分造血细胞都是以一种凝集态的方式存在,这就是造血干细胞球,而这种凝集态的改变是以作为细胞微环境的细胞外基质的改变为前提,直到发生造血干细胞在骨髓腔中迁移并到血流中循环。同样,骨髓基质干细胞的凝集态也是以一种球状体的形式存在,只是在分布形式及动员条件与造血干细胞球有所不同,本文就干细胞在骨髓中的这种凝集态形式骨髓细胞球做一综述。 Cell differentiation is controlled by a complex interaction between cell autonomous signals and stimuli from the surrounding environment. In vivo, cells are connected to their neighbors and to the extracellular matrix forming a complex microenvironment that plays a crucial role in stem and progenitor cell survival, differentiation, proliferation and migration. Hematopoiesis takes place inside the bone marrow cavity in adults while the relationship between hematopoietic cells and their niches is highly dynamic. Under steady-state conditions in this cavity, hematopoietic cells exist in an aggregated way termed "hematopoietic stem spheroids" . Any changes about the condensation of spheroids base on the affection of extracellular matrix which result in circulation of hematopoietic cells in the bloodstream following their migration within the marrow cavity. Similarly, the aggregation of stromal stem cells also lives as spheroids, but the distribution and expression between them are different. In this review, we will highlight this kind of spheroids of stem cells within the bone marrow.
