摘要 精神活性物质滥用能引起神经细胞特定的适应性反应,从而导致依赖。同时,滥用物质本身对机体细胞也可表现出一定的毒性损伤作用。研究表明:DNA甲基化作为一种重要的表观遗传修饰作用,其对基因表达的调控可能是导致这种适应性反应及毒性损伤的重要机制之一。该文介绍了DNA甲基化的分子机制及影响因素,并以酒精、甲基苯丙胺、吗啡、可卡因为例,阐述了精神活性物质的药理学及毒理学作用与DNA甲基化之间的密切关系。 Psychoactive substances abusing may induce neuron- specific adaptation and lead to physical and psychological de- pendence. Moreover, psychoactive substances have toxicological actions to cause physical damages. It has been reported that DNA methylation,acting as an epigenetic modification, may be one of the mechanisms involved in these adaptive changes and physical damages. This review shows a general introduction to the molecular mechanism of DNA methylation, and expounds the pharmacological and toxicological effects of some psychoactive substances on DNA methylation, including ethanol, methamphetamine, morphine, and cocaine.
