Expression of TGF-β1 in frontal lobe following subarachnoid hemorrhage in rats

摘要 目的探讨蛛网膜下腔出血后大鼠额叶脑组织中转化生长因子β1表达的变化及其意义。方法枕大池2次注血法制备蛛网膜下腔出血模型。30只成年健康雄性SD大鼠,随机分为对照组(仅在蛛网膜下腔注入等量生理盐水,n=6)和SAH组(n=24),其中SAH组分别于第1、3、5、7天随机均分为4组(n=6)。通过HE染色光镜下观察基底动脉的病理学变化,测定血管腔内径,免疫组化法检测各组大鼠额叶脑组织中TGF-β1在不同时间点的表达。结果与对照组比较,SAH各组基底动脉内径明显减小(P<0.01);SAH1 d时额叶脑组织TGF-β1表达增加,3 d时达高峰,而5、7 d时低于1、3 d(P<0.01),但仍高于对照组(P<0.01)。结论SAH后额叶脑组织中TGF-β1的表达发生了变化,并与CVS有关,提示TGF-β1参与了CVS的病理过程。 Objective To investigate the expression and significance of TGF-β1 (transforming growth factor-β1 ) in frontal lobe following subaraehnoid hemorrhage (SAH) in rats. Methods Double autologus arterial blood injection through eisterna magna was underwent to estabalish SAH model in rats. 30 healthy male Sprague-dawley (SD) rats were assigned randomly into two groups: Control group(injected normal sodium through cisterna magna only, n = 6), and SAH group (n = 24). The latter group were equally divided into 4 subgroups according to time points (1 d, 3 d, 5 d, 7 d, n =4). The morphologie changes of basilar artery (BA) were examined with hemotoxylin and eosin (HE) staining, and the expression of TGF-β1 in frontal lobe were detected by immunohis-tochemistry method. Results Compared with control group, the BA inner diameter of SAH group were notably decreased (P〈 0.01). The expression of TGF-β1 incresed continuely in SAH group, and reached to the peak on the 3rd day, and deceased on the 5th and 7th day, but the expressions on 5th and 7th day were still more obervious than those of the control group(P〈0.01). Conclusions The expressions of TGF-β1 in frontal lobe after SAH have changed and been concerned with CVS suggests that TGF-β1 may patieipate the patho-proeess of vasospasm.
