肝移植自发免疫耐受大鼠移植物内Foxp3的表达

摘要 目的探讨肝移植自发免疫耐受大鼠移植物内Foxp3的表达。方法双袖套法建立大鼠原位肝脏移植模型。取肝组织,通过实时荧光定量PCR、检测大鼠移植肝内Foxp3 mRNA的表达。结果术后7 d,耐受组移植物内Foxp3 mRNA表达水平明显高于排斥组和同基因组(P<0.05),术后14 d,耐受组移植物内Foxp3 mRNA表达水平亦明显高于同基因组(P<0.05),术后28 d,耐受组与同基因组移植物内Foxp3 mRNA表达水平无明显差异(P>0.05)但仍高于正常值。结论转录因子Foxp3可能参与大鼠肝脏移植自发免疫耐受的形成。 Objective To investigate the expression of transcription factor Foxp3 in the orthotopic liver transplantation modle of inbred rats with spontaneous tolerance. Methods The orthotopic liver transplantation modle of inbred rats was established according to double-sheeve technique. The total RNA isolated from liver was reversely transcribed into cDNA. The real-time fluorescence quantitative PCR (RFQ-PCR) method was used to analyze the expression level of Foxp3 mRNA in tolerance group (TOL) and syngeneic group (SYN). The expression of Scurfin in hepatic tissue was assayed by Western blot and then analyzed with computer imaging system. Results The expression of Foxp3 mRNA was lower than normal liver in the early phase after liver transplantation. About one week later, the level of Foxp3 mRNA ascensus to peak value. Latterly, the value of Foxp3 mRNA decreased but still kept in a high level than TOL and SYN. The Western blot shows us the resemblance change about Scurfin. Conclusion Transcription factor Foxp3 may play an important role in the spontaneous tolerance in the orthotopic liver transplantation of inbred rat.
