白苏不同部位中齐墩果酸和乌索酸的测定比较 被引量:1

摘要 建立反相高效液相-光电二极管阵列检测器法(RP-HPLC-PAD)测定白苏叶、茎、根中的齐墩果酸和乌索酸,并对2种三萜酸成分进行定性定量分析。采用KromasilC18色谱柱(4.6mm×250mm,5μm),以甲醇-0.2%磷酸溶液(90:10,v/v)为流动相进行等度洗脱,流速0.8mL/min,检测波长210nm,柱温25℃。齐墩果酸在0.1248μg^2.496μg、乌索酸在0.2184μg^4.368μg范围内具有良好的线性关系,平均回收率分别为98.5%和101.3%。选定的色谱条件可精确的检测白苏不同部位中齐墩果酸和乌索酸的含量。 A determination method of oleanolic acid and ursolic acid in different parts of Perilla frutescens (L.) Britton was established by reversed-phase high performance liquid chromatography (RP-HPLC) with photodiode array detector, and the qualitative and quantitative analysis for two triterpene acids were further carded out. The method was carried on a Kromasil C18 column (4.6mm×250mm, 5μm) and equivalent elution was achieved by methanol-0.2 % phosphoric acid solution (90:10, v/v) as the mobile phase. The flow rate was 0.8 mL/min and the detective wavelength was 210 nm, the column temperature was 25℃. Under these conditions, the calibration curves ofoleanolic acid and ursolic acid were linear within the range from 0.1248μg to 2.496μg, 0.2148μg to 4.368μg, and the average recovery were 98.5% and 101.3%, respectively. The method can accurately determine the contents ofoleanolic acid and ursolic acid in different parts of P. frutescens (L.) Britton.
